NMN stands for nicotinamide mononucleotide and is a precursor to NAD+, or nicotinamide adenine dinucleotide. NMN is the direct precursor of the essential molecule nicotinamide adenine dinucleotide (NAD+) and is considered a key component to increase NAD+ levels in cells. NAD+ is a critical molecule found in every cell of your body, but levels of NAD+ naturally fall with age, making it — and NMN, as a result — crucial.

NMN is one of the most popular supplements in the anti aging community. The NAD story took off toward the end of 2013 with a high-profile paper by Harvard's David Sinclair and colleagues. Sinclair, recall, achieved fame in the mid-2000s for research on yeast and mice that suggested the red wine ingredient resveratrol mimics anti-aging effects of calorie restriction. This time his lab made headlines by reporting that the mitochondria in muscles of elderly mice were restored to a youthful state after just a week of injections with NMN (nicotinamide mononucleotide), a molecule that naturally occurs in cells and, like NR, boosts levels of NAD.

However, NMN is like a large bed that movers are trying to get through a door: It doesn’t enter the cell easily. One way for NMN to enter the cell is for it to chemically transform into another molecule (called nicotinamide riboside, or NR) before it can enter the cell. NR has earned a name for itself as a highly efficient precursor to NAD+ and can enter the cell as is. NMN, meanwhile, sometimes becomes NR before entering the cell, where it chemically transforms back to NMN and then ultimately becomes NAD+.

We've learned that NR leads to NAD+ and that it also has its own pathway that bypasses various steps other NAD+ precursors have to take. In early 2019, though, new research revealed NMN might only have to become NR for certain cell types, as NMN can enter cells in the small intestine of mice. It’s unclear whether or how this will translate to humans.

To reduce the confusion, do note that NR, nicotinamide mononucleotide (NMN), Niacin (NA) and Niacinamide (NAM) are all NAD precursors, meaning all these precursors will be transformed into NAD. That said, each precursor has it's own different pathway that will lead to NAD.


What Does NMN Supplement Do?

In numerous studies (2020), supplementation with NMN has increased NAD+ biosynthesis, suppressed age-related adipose tissue inflammation, enhanced insulin secretion and insulin action, improved mitochondrial function, improved neuronal function in the brain, and more. Here, we look at the science behind NMN, its stability, possible pharmacokinetics, transport, function, and ability to induce biosynthesis of NAD+. Supplementing NMN may be an effective nutraceutical anti-aging intervention, with beneficial effects on a wide array of physiological functions.

Can NMN truly reverse aging? In a 2013 Harvard study, the Harvard scientists observed that “NMN was able to mitigate most age-related declines in mice,” and that “treatment of old mice with NMN reversed all of these biochemical aspects of aging.” The study concluded that “enhancing NAD+ biosynthesis by using NAD+ intermediates, such as NMN and NR (Nicotinamide Riboside), is expected to ameliorate age-associated physiological decline”.

Chromadex NMN Supplement Test Report: 64% of NMN Supplements Fail Tests

ChromaDex NMN Report (Oct 2021): Supplement manufacturer ChromaDex alleges that testing has revealed that more than half of NMN anti aging supplements being sold on Amazon contained almost none of the bioactive molecule.

ChromaDex markets TruNiagen, which is based on its branded Niagen ingredient, which is a form of nicotinamide riboside (NR), the form of nicotinamide that actively crosses into the cell to participate in the NAD+ pathway. Boosting this pathway, and thus maintaining cellular function at a lively, youthful level, has become a prominent theme in recent anti aging research.

Some researchers have argued that a transporter exists for NMN too, which is something that ChromaDex disputes. It claims that NMN must first be converted into NR in the blood before it can be used by the cells.

In the several years since the NMN transporter discovery was asserted, the NAD+ anti aging field has become crowded with NMN based supplements all seeking a piece of the pie created by ChromaDex and its competitor and one time customer Elysium. (The two companies have been locked in various patent infringement and breach of contract suits for several years.)

Last fall, ChromaDex obtained 22 samples of NMN products being sold on Amazon. Of these, 14 claimed to contain 500 mg of NMN. A further two products claimed 300 mg on the label; five claimed 250 mg and one product claimed 125 mg.

Only one of the 500 mg products came close to meeting label claim. It had 456 mg of NMN. All of the other 13 products in the 500 mg group either came in at below reporting limit (BRL, defined as less than 1% of the stated amount) or, in the case of 3 products, no NMN was detected at all.

Almost all of the rest of the products performed better, having at least 88% of the label claim. A lone 250 mg product was identified as BRL.

In summary, ChromaDex said that 64% of the products tested contained less than 1% of the stated amount of the active ingredient, which should give consumers pause, the company said.

"While this is a limited snapshot of the vast NMN finished product landscape, it does provide a glimpse into the high variability of product quality that is available... According to this study, the majority of the products one might purchase online contain such a small amount of NMN that there would be no clinical benefits achieved from the dose. Another concern with these adulterated products is that the actual contents are not known and could pose a risk to the user," the company said in a statement.

Editor's Note: In February 2022, another third-party investigation uncovered tens of thousands of fake supplements sold on Amazon. Many of these included NMN.

David Sinclair Supplements List

Keep in mind that to date, Dr. David Sinclair has made a point to not promote or endorse any supplement products. We compiled this list from interviews and books in which Professor David Sinclair mentions supplements he takes.

We don’t know if he still takes these supplements, or whether he takes additional supplements that are not included on this list.

For example, in his book Lifespan, he mentions on page 304 that he takes NMN and resveratrol every morning, often mixed in his yogurt.

Here is the summary list:
  1. Nicotinamide Mononucleotide (NMN), 1g per day, in the morning
  2. Quercetin and fisetin, 500 mg each, once per day, in the morning
  3. Resveratrol, 1g per day, in the morning
  4. Vitamin D3 and K2
  5. Spermidine, 1 mg per day, in the morning
  6. Metformin (prescription drug) 1 gram per day: 500 mg in the morning and 500 mg in the evening
  7. Statin (prescription drug) – taken since his early 20s due to family history of cardiovascular disease
  8. Low-dose aspirin – 83 mg per day
  9. Alpha lipoic Acid
  10. Coenzyme Q10

NR vs NMN Molecule

Pitting NR vs NMN against each other is, for now, somewhat of a moot point because the two molecules have never been studied side by side in humans. The biggest, and most obvious, difference between NMN and NR is size. NMN is simply larger than NR, meaning it often needs to be broken down to fit into the cell. NR (Nicotinamide Riboside), when compared to other NAD+ precursors (like nicotinic acid or nicotinamide) reigns supreme in efficiency.

But give NMN a new door, one it can fit through, and it’s a whole new game. This is where cellular transporters come into play. Transporters are proteins that are doors on the cell; they allow molecules to enter the cell without needing to chemically transform.

Research from Shin-ichiro Imai, M.D., Ph.D., a professor of developmental biology at Washington University in St. Louis, identified a transporter that allows NMN to get into the cell without converting to NR. The catch? The transporter is only on cells located in the gut of mice and only works in the presence of sodium ions. NR, however, has been shown to enter cells in the liver, muscle, and brain tissue of mouse models. To date there is not yet evidence that these mouse studies can be extrapolated to humans. But again, the two have never been matched up against each other in a way that can truly identify one as superior to the other.

As research grows, it is likely to become clearer that NMN and NR may have differing benefits depending on the part of the body and their use. Where NMN transporters are found, they could make one more preferable over the other. NMN has also been found to improve insulin activity and production which could help to accelerate metabolisms and make the body more glucose tolerant.

Stability and storage of NMN and NR

A stability analysis demonstrates NR is stable (does not undergo degradation) for up to six hours at room temperature and seven days at two to eight degrees Celsius. Regarding stability of NMN and NR, David Sinclair, PhD, A.O. of Harvard University says, “Make sure your NR and NMN is kept in the cold. If it’s just on the shelf and it’s not in a stabilized form, then it will degrade to nicotinamide, which is something you don’t want to take high doses of because we’ve shown in my lab many years ago that nicotinamide will inhibit the sirtuins, and PARP as well, and interfere with DNA repair.”

What Foods Contain NMN?

NMN (Nicotinamide MonoNucleotide) is a molecule found in various foods such as broccoli, cabbage, avocado, mushrooms, meat, and shrimp. However, obtaining sufficient amounts through diet alone can be challenging. 

Studies suggest that daily dosages of NMN range from 50mg to 250mg, and a 150-pound (68kg) person would require approximately 560mg per day. Unfortunately, obtaining these amounts solely through diet would be impractical. For example, you would need to consume about 100 pounds of edamame, 1,800 pounds of broccoli, or unrealistic amounts of cucumber, cabbage, avocado, tomato, mushrooms, raw beef, or shrimp to achieve the required intake. Therefore, taking NMN supplements may be a more practical approach to ensure adequate daily intake.

NMN Supplement dosage and side effects?

NMN is an ideal supplement to increase cellular levels of NAD because it’s well-tolerated, and both human and animal studies have observed it had minimal side effects.

A clinical study (GeroScience 2022) suggests that NMN supplementation can indeed elevate NAD+ levels and increase walking distance in healthy participants, and has narrowed in on 600 milligrams a day being the optimal dose.

Research in humans has shown that doses of up to 1,200 mg daily are safe to consume.
